Humanin (HN) exert s a cyt oprot ect ive act ion in t he presence of pro-oxidat iveagent s in several t issues. Recent ly, we demonst rat ed t he expression of HN in t heovary of prepuberal and adult rat s. Also, we showed a different ial locat iondependent on t he follicular st age. In t he present st udy, our aim was t o evaluat et he ant i-apopt ot ic act ion of HN, as a cyt oprot ect ive mechanism against oxidat ivest ress in granulosa cells. To explore t his aim, we examined t he effect of HN onapopt osis in granulosa cells from ovaries of prepuberal and adult rat s incubat ed ina pro-oxidat ive environment . Thus, we incubat ed each ovary from prepuberal oradult rat s wit h HN (1μM) for 30 min and H2O2 (150 μM) for furt her 1 or 2 h,respect ively. Cont ralat eral ovaries from rat s of each t reatment were used asrespect ive cont rols. We det ermined t he number of TUNEL posit ive granulosa cellsper follicle.
