Cytogenetic and Molecular studies of a population of Boana pulchella from the Dique Cruz de Piedra (San Luis province) and Molfesa (Buenos Aires province) Argentina

Resumen:
The species Boana pulchella is widely distributed along the countries of Brasil, Uruguay y Argentina. In this work were characterized cytogenetically and molecularly, two populations of this species located in Dique Cruz de Piedra (San Luis province) and Molfesa (Buenos Aires province). Both populations were subjeted to the techniques of Giemsa staining, Silver staining, Constitutive heterochromatin, Telomeric fluorescent ?in situ? hibridization (telomeric FISH) and Sequencing of the enzyme COI. Ten male adult specimens were analyzed, whose karyotypes were obtained with the culture of peripheral blood lymphocytes whithout the sacrifice of the specimens. Both populations presented a 2n=24 and NF=48; The chromosome formula of the San Luis population was 5m+7sm and Buenos Aires population was 4m+8sm; Silver staining of both populations showed marks at the satellites of the pair 12; Constitutive heterochromatin showed that both populations share the same pattern at the pairs 1, 2, 4, 5, 8 y 12 and differed at the pairs 3, 6, 7, 9, 10 y 11; Telomeric FISH showed marks at all pairs in telomeric sites of both populations; Sequencing of the enzyme COI determined that the specimens of both populations belong to Boana pulchella and that there were differences between the populations studied in this work and the populations of Brasil (data provided by UNESP of Brasil), but not enought to separate them as a species. So we concluded that the specimens of San Luis and Buenos Aires belong to Boana pulchella but they represent different geografhical variants of this species.
