In addition to the epigenetic and genetic changes that occur in epithelial cells, in recent years it has been shown that tumor progression also depends on the bidirectional dialogue between tumor epithelial cells and surrounding stromal cells. We recently demonstrated that human renal adipose tissue (AT) surrounding a tumor regulates the behavior of tumor and non-tumor renal epithelial cells, differently from normal AT. In this work we asses the effect of soluble factors secreted by tumor and non-tumor renal epithelial cells on perirenal AT fragments from healthy patients. We obtained conditioned media (CMs) from tumor (786-O, ACHN and Caki-1) and non-tumor (HK-2) human renal epithelial cell lines and incubated AT fragments with these MCs for 48 h. Then, part of the fragments was lysed and total proteins were obtained; and part of the fragments was embedded in paraffin for histological analysis. In the lysis and/or histological sections we evaluate changes in the expression of: 1) Perilipin A (mature adipocytes marker); 2) UCP1, HSL, TBX1, cEBP alpha, PGC1 alpha (brown/beige adipose tissue markers); 3) leptina, adiponectina and ObR.
