SERUM SIALYLATED N-GLYCANS AS POTENTIAL BIOMARKERS FOR PROGNOSIS IN CHRONIC CHAGAS' DISEASE

Trypanosoma cruzi, the causative agent of Chagas Disease, affects millions of people worldwide, especially in Latin America. Chronic infection can evolve to cardiomyopathy and/or digestive abnormalities in up to 30% of infected patients. The aim of this work was to explore if patients with the asymptomatic and cardiac forms present changes in their serum N-glycome profile, potentially useful as prognostic markers in the chronic phase of this disease. For this, N-glycans released from sera of asymptomatic (n=9) and cardiac patients (n=7) as well as non-infected individuals (n=8) were characterized both by hydrophilic interaction liquid chromatography (HILIC-UPLC) and weak anion exchange chromatography (WAX-HPLC). Our results showed that cardiac patients show an increase in trisialylated structures (A3G3S3, FA3G3S3 and A4G4S3) in comparison with asymptomatic and non-infected individuals, while tetrasialylated structures are diminished in infected individuals compared to non-infected subjects. Interestingly, a negative correlation was observed between tetrasialylated species with parasite load measured in human blood samples from both asymptomatic and cardiac patients by qPCR assays. Our preliminary research suggests that the serum N-glycome of the host is altered by the disease and demonstrates its potential as source of prognosis biomarkers.
