Resumen:
Sociedad chilena A high percentage of therapeutic failure with meglumine antimoniate (MA) has been observed in patients from Salta in the last years. The aim of this study was to assess the association between treatment failure and sensitivity to MA from clinical isolates of Leishmania sp. A total of 20 clinical isolates were included in this study and data of clinical form and treatment response of patients to MA were collected from their medical records. Isolates were obtained by needle aspiration from the border of cutaneous (CL) and mucocutaneous(ML) lesions. Isolated parasites were identified by PS-PCR (polymorphism specific PCR). The MA sensitivity assay was performed exposing 106 promastigotes/mL from each isolate to different concentrations of MA (0.8-40 mg/ml). Leishmania (V) braziliensis MHOM/BR/75/M2903 strain, which is known assensitive to MA, was used as a control. Each assay was carried out in triplicate and the promastigotes were counted in the Neubauer chamber. The IC50 was calculated for each isolate and resistance phenotypes were determined by Kruskal Wallis test using GraphPad Prism 6.01. Categorical variables were analysed by Fisher test with SPSS 2.0. Among the analysed patients, 70% exhibited (CL) and 30% (ML). All the isolates were identified as Leishmania Viannia, 35% (N=7) showed an IC50 significantly higher than the IC50 of M2903 (p
