Resumen:
As intracelullar viscosity might reach values of about hundreads of centiPoise (cP) it is interesting to study its effects on protein dynamics and its influence on electron transfer activity. To this end we study the two types of copper metalloproteins adsorbed onto gold electrodes coated with self-assembled monolayers (SAMs) immersed into ionic liquid/water blends of different compositions, reaching viscosities of about 1000cP. Two types of copper sites were chosen, mononuclear T1 copper sites from Pseudomonas aeruginosas´ Azurin and the binuclear CuA site from Thermus thermophilus. The objective of this works is to assess the influence of viscosity on the main ET parameters, altering protein dynamics in order to understand the underlying ET mechanism.
