Type IV secretion Systems (T4SSs) are bacterial nanomachines thattransfer proteins across the bacterial cell envelope and into target cells. Wehave recently provided the first demonstration that a T4SS mediates thecontact-dependent killing of other bacterial cells. One Xanthomonas T4SS effector XAC2609, whose gene islocated in this special T4SS operon, is secreted into target cells. It has invitro lysozyme-like lytic activity and is inhibited by the soluble domainof a predicted lipoprotein called Xanthomonas T4SS immunityprotein XAC2610. Here, we set out to investigate the in vivo function of XAC2610. Since XAC2609 ispredicted to act in the bacterial periplasm and XAC2610 has a lipobox which directs it to the bacterial outer membrane, the hypothesis is that XAC2610 acts as an immunity factor against the possiblydetrimental autolytic effects of XAC2609.
