Cattle undergo numerous environmental and management stressors, which reduce fertility and affect ovulation. The extracellular matrix of the ovarian follicular wall can be altered by matrix metalloproteinases (MMPs), whose activities are regulated by interleukins and tissue inhibitors (TIMPs), especially during ovulation. The aims of the present study were to evaluate changes in the hormonal milieu as well as the localization and activity of MMP2 and MMP9 and the localization of MMP14, TIMP1 and TIMP2 in response to ACTH in the preovulatory period in cows, and to determine the direct effect of ACTH on the mRNA expression of MMP2 and MMP9 in the cultured follicular wall of bovine ovaries obtained from the abattoir. ACTH was administered during proestrus every 12 hours until ovariectomy, which was performed before ovulation.
