ObjectivesDecidualized cells sense embryo quality allowing the implantation of normal development (ND) blastocysts while preventing the invasion of impaired development (ID) ones. Understanding the molecular basis of this process may contribute to improve infertility treatments. We aim to characterize the immune microenvironment generated by decidualized cells in response to soluble factors secreted by ND and ID blastocysts.MethodsHuman endometrial stromal cell line (HESC) was in vitro decidualized with medroxiprogesterone and dbcAMP during 8 days. Then, decidualized cells (dHESC) were stimulated with blastocyst conditioned media (BCM). BCM were recovered from 5 days individually cultured blastocyst obtained from IVF or ICSI and classified as ND or ID according to morphological standards. After 24h BCM stimulation, dHESC gene expression (RT-qPCR) and protein production (flow citometry) were evaluated. Supernatants from 48h stimulation were used to test Tregs migration (transwell system) and during dendritic cells (DCs) differentiation. For statistical analysis, p
