Background Progesterone is a fundamental hormone in the generation of the materno-placental interface while vasoactive intestinal peptide (VIP), produced by trophoblast and immune cells, modulates the maternal immune response towards a tolerogenic profile. Here, we characterized the relation between the endogenous production of VIP by endometrial stromal cells (HESC line) modulated by progesterone and its effect on RANTES expression. Methods We evaluated the expression of RANTES and KLF13 on HESC cells by RTqPCR under different combinations of LPS, progesterone, VIP and VIP-antagonist. Results LPS stimuli induced RANTES while its combination with progesterone or VIP potentiated this effect. KLF13 was induced by progesterone and VIP and LPS enhanced it even more. VIP-antagonist partially reversed the effect of progesterone in both cases. Conclusions The effect of progesterone on RANTES is partially mediated by VIP and KLF13 is probably involved in this mechanism, however, KLF13 expression is not enough to induce RANTES.
