Lavandin essential oil combined with the biopolymer PHBV, poly (3-hydroxybutyrate-co-3- hydroxyvalerate), for wounds

In this work, poly (3-hydroxybutyrate-co-3-hydroxyvalerate) PHBV was combined with lavendin essential oil (LavEO), from Lavandula hybrida growth in Argentina. Linalool (36%), linalool-acetate (29%) 1,8-cineole (6%) and camphor (6%) were its main compounds, analysed here by CG-MS.PHBV is a biodegradable copolyester from bacterial source; PHBV porous scaffolds have shown to be suitable for fibroblasts and keratinocytes proliferation [1] [2] and thus for skin regeneration. In addition, LavEO exhibits strong anti-inflammatory and antibacterial activity, what improves wound healing [4][5]. Purified monoterpens as well as complete EOs have been combined with other biopolymers for different applications, but scarcely for wound healing and not yet with PHBV [6]. 100-200 µm thickness PHBV porous membranes containing LavEO at 2, 4 or 8% m/m were obtained by emulsion solvent evaporation method. LavEO-PHBV membranes did not shown cytotoxicity when tested with NIH/3T3 fibroblasts (according to the ISO 10993-5 standard). Membrane analysis by SEM showed in all cases a high level of porosity, ̴ 20% in surface. Keratinocytes (KC) adhesion and proliferation were evaluated with cells of HaCaT line. Membranes made with LavEO 2 or 4 % m/m, allowed KC proliferation at the same level of PHBV membrane controls. However, in those with LavEO 8% m/m, cells neither adhere nor proliferate, even if their hydrophobicity, estimated by contact angle, was similar to LavEO 4%. The reduction in the elastic modulus determined by DMA due to the presence of LavEO remarks its plasticizer effect (Control E=305±10 MPa; LavEO 8% E=150±20 MPa). According to these results, LavEO-PHBV 2 or 4% membranes become a promising solution specially for infected and chronic wounds, often arrested at the inflammatory phase.
