Kidney acellular matrix and new strategies of recellularization

Replacement therapy is the best treatment option againstorgan failure, however graft rejection and adverse effects of immunosuppressionremain major clinical complications. The aim ofthis work was to obtain kidney acellular matrix and develop newstrategies of recellularization.Wistar rats were used, which weremade nephrectomy, then the kidney was cannulated for renalartery, placed in a bioreactor at room temperature and connectedto a pump with a constant flow of 0.5 ml/min for the passage of1% SDS and distilled water (18 h). Macroscopic changes in thekidney during the decellularization can be displayed in our video (https://youtu.be/NI2OU-PE15g) . In histological sections an acellularmatrix was observed with preserved renal architecture beingpositive for markers of collagen and laminin. For recellularizationprocess we develop an in vitro and another in vivo protocol. In thefirst, 1.5 x 106 human renal epithelial cells (HK-2) was administeredby ureter. At 72 hours, with histological techniques viable cellsmainly located in the cortical tubules were observed. For in vivoprotocol, acellular scaffold were implanted into recipient rats insubcutaneousand intraperitoneal location for one or two weeks.Once recovered the scaffolds showed a large inflammatory infiltrate,along with some positive cells mar..ers for transgenlin andpodocin. Conclusion: These results show the feasibility of renaldecellularization techniques for obtaining acellular scafolds, andthe possibility of recellularize at least partially both in vitro and invivo. However, the development of more complex recelullarizationtechniques is required to generate more complex and functionalrenal structures.
