Technicaland experimental advances in DNA amplification, quantitative PCR, cDNAmicroarray and chromatin immunoprecipitation (ChIP) analysis have ledto an increase in the number of studies identifying specific genesregulated by chromatin related proteins. Our aim was to characterizeactivated/repressed genes in response to nicotine when chromatinremodeling is involved. We decided to setup a genome screening methodwhich sequentially combines two key techniques: 1) the ChIP assay,used to study protein-DNA interactions and mapping of histonemodifications, 2) Random amplified polymorphic DNA (RAPD), which hasbeen used to detect polymorphism in genetic mapping. To approach thisstrategy, we chose a simple nicotine responder model, PC12 cells. NGF,the strongest PC12 stimulus, was used to setup the technicalconditions. Different antibodies against histone modifications weretested for ChIP. For RAPD, around 10 randomized primers were used tofind different unknown amplicons. Preliminary results indicate thatspecific genes were detected with different antibodies. Sequentiationof the relevant bands generated with RAPD will allow us to identifythese genes.
