A PRELIMINARY LINKAGE MAP IN PEA (Pisum sativum L.) BASED ON SRAP MARKERS (SEQUENCE-RELATED AMPLIFIED POLYMORPHISM)

A mapping population of Pea was derived from a cross between thecultivars Zav 25 and DDR11 with a view to generating a new geneticlinkage map of Pisum sativum L. based on SRAP markers. Fifteenprimer SRAP combinations were used to detect 183 polymorphicbands (12,2 polymorphic bands/primer combination). The mostpolymorphic primer combination was Me3-Em2, with 29polymorphic bands. Fragments generated by SRAP were treated asdominant markers. Goodness-of-fit between the observed andexpected segregation ratios was determinate by a Chi-square test.Markers without a significant level of segregation distortion (χ2>χ2α=0.01) were used to construct the map on JOIN MAP v4 software.The linkage map consists in 50 genetic markers distributed on 7linkage groups (LGs) covering a total of 310,5 cM. The LGs lengthranging from 20.4 to 80.8 cM (mean 44,29 cM) and including 4-10markers.
