Introduction: Previously, we showed that IGF2BP1 knockdown (KD) leads to a decrease in ABCB1 and ABCC3 expression (proteinand mRNA). Sorafenib (SFB) treatment induced IGF2BP1, ABCB1 and ABCC3 expression and KD of IGF2BP1 prevented theinduction of ABCB1 expression, ABCC3 induction was prevented only at the mRNA level. Objective: To study the molecularmechanism of ABC transporter regulation by IGF2BP1 and its influence on SFB chemotherapy. Methods: KD of IGF2BP1 wasdone by transfection of a siRNA (IGF2BP1 siRNA), scrambled (C siRNA) was used as control, in HepG2 cells. After 24 h oftransfection, cells were treated with the transcription inhibitor Actinomycin D (ActD) 5 ug/ml for 6 h. Also, HepG2 cells werepre-treated with ActD 5 ug/ml for 30 min and incubated with SFB 2µM for 48 h. Control cells received DMSO. IGF2BP1, ABCB1and ABCC3 mRNA levels were measured by Real Time PCR.
