Resumen:
The non obese diabetic (NOD) mouse develops a spontaneous Th1 response against exocrine glands that resembles Sjögren?s syndrome. VIP is a neuroimmuno peptide mediating secretory and anti-inflammatory responses promoting a Th2 profile. Similarly, pregnancy has been described as an anti-inflammatory status where uterus quiescence and predominant Th2 response are observed. The aim of the present study was to investigate NOD macrophages profile and VIP anti-inflammatory effect in both pregnant and non-pregnant mice. We present evidence of a higher proinflammatory response (IL-12, nitrites, TNF-a) and lower IL-10 production of NOD macrophages to LPS that was reversed by VIP in a concentration-dependent manner and through the production of IL-10. Pregnancy normalized macrophage profile of NOD mice. Our results show an anti-inflammatory effect of VIP on NOD macrophages that is comparable to that of pregnancy in lowering Th1 and stimulating Th2 cytokines
