Focal Adhesion Kinase (FAK) is a cytoplasmic protein kinase of 125-kDa implicated in several cellular functions. This protein is an attractive molecular target for cancer therapy due to a wide variety of studies that have demonstrated associations between the activation and higher expression of focal adhesion kinase (FAK) with tumor progression, invasion, and drug resistance in malignant tumors. This work is a review of the evaluated strategies to inhibit the FAK activity on solid tumors. Also, this review includes an overview of the preclinical (in vitro and in vivo) and clinical studies of FAK inhibitors.
