Ferroportin expression in anemic mice

Ferroportin (MTP1) is the sole iron exporter identified in mammals that participates in iron release from both enterocytes and macrophages. We studied the cellular localization of MTP1 in duodenum and spleen of anemic and no anemic mice. Hemolytic anemia was induced in mice using phenilhydrazine (60 mg/kg).Tissues were removed and fixed with formol 10%. Sections were processed for immunohistochemistry using a primary antibody (1:1000) and the Envision+System-HRP(DAB) kit. Perls´ iron staining was performed. No anemic mice showed cytoplasmatic expression of MTP1 in enterocytes, while in anemic mice was detected at the basal membrane. Macrophages of the red pulp in no anemic mice showed co-localization of MTP1 with hemosiderin. In spleen of anemic mice MTP1 was not observed. Anemia induces translocation of MTP1 to basal membrane of enterocytes, without splenic changes. We show tissue specific changes in MTP1 expression in hemolytic anemia
