It is well known that duodenal expression of Divalent Metal Transporter 1 (DMT1) depends on dietary iron. However, little is known about the regulatory relationship between this iron transporter and the erythroid regulation. We studied duodenal DMT1 expression under hemolytic anemia condition. Mice were treated with phenylhydrazine (60 mg/kg) (days 0 and 2) in order to induce anemia. Control mice received saline solution. Expression of DMT1 in duodenal tissue was evidenced on days 3, 4 and 5 using both specific affinity purified polyclonal anti-DMT1 antibody and the Envision+ System-HRP(DAB) kit. Control mice showed intracellular DMT1 expression in enterocytes, while in anemic mice both intracellular and apical DMT1 expression was detected on day 5. We concluded that hemolytic anemia induced a translocation of DMT1 expression from intracellular compartment to the brush border membrane of the enterocytes. These results suggested that duodenal DMT1 expression may be regulated by erythroid regulator.
