Soybean is the main crop of Argentine. In spite of high nutritional value, soybean and foods containing soybean are repelled for consumers, because oxidation of polyunsaturated fatty acids by isozymas lipoxygenases produces beany off-flavors. Soybean seeds contain three isozymes controlled for three dominants genes Lx1, Lx2 and Lx3. Among the three isozymes, Lox2 largely contributes to the generation of a beany off-flavour. Mutant germoplasm Lx1, Lx2, Lx3 and Lx1Lx2Lx3 have been identified. Molecular markers are tools that allow the quick and efficient incorporation of mutations to adapted germoplasm at the breeding programs. This work proposes to validation of molecular markers to select Lx2 mutants in soybean. A F2 population was obtained for crossing between a cultivar and a genotype lacking three lipoxygenases. Gene Lx2 segregation was studied through use two molecular markers, one type PCR-RFLP and one type SSR, and phenotypicaly through method SDS-PAGE. Mendelian segregation was expected for molecular markers, and association between molecular results and phenotypic analysis was present. Both molecular markers were efficient to selection Lx2 mutant.
