Facultad de Medicina, UBA), Bs As, Argentina.Background: We observed that lysophosphatidic acid (LPA), through its receptor LPA3, regulates pivotal mediators of blastocyst adhesion and invasion. So, our aim was to investigate the role of LPA3 receptor in the process of embryo implantation. Methods: Wistar females on day 5 of gestation were injected with an LPA3 antagonist (DGPP, 0.1 mg/kg, intra-uterine). Fetoplacental units, placentas and uteri were macro and microscopically (hematoxilin & eosin) evaluated on days 15 and 21 of pregnancy. Results: DGPP caused embryo resorption (70±5%) and aberrant spacing. Resorpted sites presented a reduction in the decidua (p<0.001) and fetoplacental weight (p<0.001), as well as anomalies in their histological architecture. An increase in erythrocyte extravasation, neutrophils infiltration, fibrinolysis and signs of cellular death in placental cells were observed. However, uteri weight and histology was not modified. Conclusions: LPA3 signalling during the window of implantation modulates crucial later events as decidualization and placentation, thus affecting pregnancy outcome.Key words: implantation; decidualization; placentation; LPA3 receptor.
