Neurons arehighly polarized cells typically extending several short, thick taperingdendrites and one functionally distinct long thin axon, which differnot only in their morphology but also in membraneproteins, organelles. In pyramidal neurons, Golgi apparatushas a polarized distribution that plays an important role in the developmentand maintenance of neuronal polarity, that is complemented bysatellite Golgi outposts (GOPs) in dendrites, which play avital role in synaptic plasticity and dendrite development. However, manyquestions remain about cellular mechanisms that regulate thisspatial distribution. MAP6d1 is a protein which expression is highly regulated in differentiated neurons, thatcontains two functional domains. The amino terminaldomain can interact with Golgi membranes throughpalmitoylation and the Mn module with the ability to interact withmicrotubules. Incultured neurons MAP6d1 localizes at the somatic Golgi apparatus and GOPs. Wefound by two hybrid screen that MAP6d1 interact with Tctex, a dyneinlight chain, which binds cargos to the motorcomplex. Here we shows that MAP6d1alters the morphology and polarization of neuronal Golgiapparatus and GOPs in the dendritic compartment. Ourobservations suggest that MAP6d1 could act as a regulator of the organizationand positioning of Golgi apparatus in pyramidal neurons, a phenomenon that would be of great importance togenerate and maintain the dendritic arbor and neuronal polarity.
