Modulation of the immune response induced by a genetic vaccine against Bovine Herpes Virus (BHV-1) using different adyuvants

It is well documented that adjuvants improve the immune response generated by traditional viral vaccines, but less is known about their effects on the immune response elicited by DNA vaccines. In this study, we have investigated the use of Montanide ISA 25, ISA 206 and IMS 12802 (SEPPIC) as adjuvants and analysed the humoral and cellular specific immune responses elicited by DNA vaccines based on the BHV-1 glycoprotein D (gD) in pCI Neo vector. The comparison of the antibody response induced in mice by DNA truncated gD (secreted version) formulated with or without ISA 25 or ISA 206 showed that the immunomodulator affects the total specific humoral response (mainly ISA 25). The isotypes induced by these adjuvants were of the type Th1/Th2. The cellular immune response induced in mice immunized with the genetic vaccine and adyuvants has been also studied. When total spleen cells were marked with specific monoclonal antibodies against surface markers, a significant increase in the macrophage and CD4+ populations of the groups receiving pCI Neo+ ISA 25 or pCI Neo+ISA 206 in comparation with PCI Neo group was observed. Our results indicate that the incorporation of ISA 25 or ISA 206 into DNA vaccines induces an increase of the humoral and cellular specific immune responces in mice.
