γδ T cells constitute a functionally specialized subset of T lymphocytes which act as early sensors of cellular stress and infection. The hemolytic uremic syndrome (HUS) associated with diarrhea, a consequence of Shiga toxin (Stx)-producing Escherichia coli infection, is a common cause of pediatric acute renal failure in Argentina. Stx type 2 (Stx2) causes direct damage to renal cells and induces local inflammatory responses which involved secretion of inflammatory mediators and the recruitment of innate immune cells. Previously, we demonstrated that γδ T lymphocytes reduced the viability of human glomerular endothelial cells (HGEC) treated with Stx2. In this work, we aimed to investigate the mechanism by which γδ T cells prompt the mortality of HGEC. To this purpose, we analyzed by flow cytometry, the expression of perforin in γδ T cells after incubation with supernatants obtained from HGEC treated with Stx2 (0.001-1 ng/ml). Also, we evaluated by confocal microscopy, the interaction between γδ T cells and HGECtreated or not with Stx2 (0.01 ng/ml).
