Resumen:
Glycoproteomics emerge as the fusion field between proteomics and glycomics, pursuing the study of biological systems at adefined condition, handling the analysis of glycoproteins, including glycan structures, protein identity, glycosylation sites andlevel of occupancy. Our group has been focused on the structural study of glycoconjugates from unicellular to higher organismwith the aim of revealing biological processes and functionality. The first steps on our research were supported with traditionalchemical and enzymatic methodology and tools like HPAEC-PAD and MALDI-TOF MS. Lately, the acquisition of a nanoHPLCESI-Orbitrap, enabled us to speed up deeper glycoproteomics approaches from single purified proteins to samples that are morecomplex. I will show some of our recent studies on the structural determination of glycoproteins obtained from different strainsof bacteria. This type of analysis allows us to find out in a purified protein both N- and O- glycosylation along with their respectiveglycan structures. I will focus in the workflow used to achieve our research for example the analysis in the positive and negativepolarities to assure the assignments. I will also show that sugar components that have not been expected may be detected inthis type of analysis and confirmed after nanoHPLC-ESI-Orbitrap analysis of the released oligosaccharide.
