The aim of this work was to identify and characterize new viruses infectinggarlic in Argentina. In 2013 samples were analyzed garlic (cultivar Mesidrome) byimmunosorbent electronmicroscopy and decoration with antisera pull: Onion yellowdwarf virus, Leek yellow stripe virus, Garlic common latent virus, Shallot latent virus,Garlic virus C and Garlic virus A. Were observed a decorated viral particles and otherundecorated. Total RNA were extracted and sent for sequencing (Illumina HiSeq 1500).The contigs were compared by Blastn with the GenBank database and showed thepresence of a contig (8336 nucleotides, nt) with 98% identity (59% coverage) with thelongest sequence GarV-B known to date (JN019813, 4923nt). The identity of nt andamino acid (aa) of the gene encoding the coat protein (CP) with others GarV-B isolateswas calculated using Lasergene 8.0.2 software package (DNASTAR, Inc., Madison,WI, USA) and showed higher values of 88.4% and 94.7%, respectively. According tothe International Committee on Taxonomy of Viruses (ICTV) criteria for differentiationof Allexivirus species (more than about 72% nt identity or 80% aa identity betweentheir CP or polymerase gene are the same species), the contig was considered the firstcomplete sequence of GarV-B (NC_025789/KM379144). The identity (nt and aa) of thepolymerase and CP available sequences of GarV-B with the others Allexiviruspublished in GenBank were calculated. The identity found was higher than thoseestablished by the ICTV in all comparisons (polymerase and CP) among isolates GarVBand GarV-X. These results suggest that GarV-B and GarV-X are the same viruses.Keywords: Garlic, Allium, Allexivirus.Work supported by INTA and CONICET.
