Comparative genomics of Acinetobacter baumannii international clone 1 reveals a high degree of synteny and gene sequence conservation

Resumen:
Acinetobacterbaumannii is anosocomial pathogen with high genomic plasticity involved in the development ofextreme and pandrugresistance. The international clone 1(IC1) has shown to have globaldissemination. We performed whole-genome comparison (WGC) studies to gaininsights into the genetic features shared by strains belonging to IC1 fromdifferent geographical regions. The strain A144 (1994, Argentina), was used asthe reference IC1 strain in the genomic studies using RAST, ACT, MAUVE, IslandViewer, PHAST, ISFinder, Geneious andHarvest software. A144 genome has 99% average nucleotide identity with A155isolated from the same hospital. Moreover, these genomes have 97% identity withAYE, AB307-0294, AB5075-UW, AB0057, A1 and D36 (IC1), 80% identity with A118(sporadic clone), 80% with ACICU (IC2), and 79% with ATCC 17978 (sporadicclone). Besides the presence of 4 inversions, 378 insertions, and 433 deletionsfound when the A144 genome was compared with the IC1 genomes, high degree of synteny wasobserved among the IC1 strains analyzed. The number of single nucleotidepolymorphisms (SNPs) between A144 and A155 were 398, and 16337 with D36,(minimum and maximum observed, respectively). With Island Viewer we identified9 regions of genome plasticity (RGP) within A144 genome that were also found inAYE. Seven out of 9 RGPs have phage related elements, denoting foreign DNAacquisition. A novel AbaR-likeisland was also identified in A144. In summary, besides the great syntenyfound among IC1, unique genomic features where observed in A144 reinforcing theidea of high plasticity of A. baumannii?sgenomes.
