DEFICIENT LIF PRODUCTION BY ENDOMETRIAL INFILTRATED CD3 T-CELLS COULD BE INVOLVED RESPONSIBLE OF RECURRENT SPONTANEOUS ABORTIONS

Leukemia inhibitory factor is essential for embryo implantation and a Th2 immune response at the fetal-maternal interface contribute to successful pregnancy. We investigate LIF expression and production by T cells in-patients with recurrent spontaneous abortion (RSA) and in-vitro fertilization failures and its immunomodulation after immunotherapy with paternal leukocytes. Methods: Endometrial samples and peripheral blood were obtained from the mentioned patients and fertile women. Samples were processed for LIF expression by Western Blot and flow cytometry. For specific LIF detection in CD3 T cells, samples were stained using mAbs anti CD3, anti LIF-Receptor and anti LIF. LIF production also was evaluated after 24h cultured in the presence of alloantigens and TNFα. Results: RSA and FIV-patients did not showed LIF expression by Western Blot. In fact, specifically endometrial and peripheral T cells from RSA (n=5) and FIV-failures (n=5) neither produced LIF detected by intracellular stained and analyzed by flow cytometry in opposite of fertile women (9%). However, endometrial CD3 T cells from RSA patients increased intracellular LIF production after treatment with paternal leukocyte (5%). On the other hand, stimulation with TNFα or alloantigens could not increased LIF production by endometrial and peripheral blood T cells. Conclusions: The defective production of LIF by infiltrated endometrial CD3 T-cells may contribute to the development of recurrent spontaneous abortions and in vitro FIV failure. This phenomenon could be reverting with allotreatment with paternal leukocytes increasing LIF production and successful pregnancy rate.
