Resumen:
When analyzing leptin promoter there were found several potential binding sites for Sp1. One of them is present inside the region identificated as PLE (enhancer of the placental leptin) and it could regulate the expression of leptin at the placental cells. In this region it is present the vetors p1842 and p1860 which conteins minimun regions from the promoter of leptin that response to E2, so it could be possible that Sp1 interact with estrogen receptors regulating the expression of the placental leptin.Sp1 is a ubiquitous transcription factor already described in the leptin gen that is involved at the activation the MAPK way.In first place, Bewo Cells where transiently transfected with the reporter vector.It was used the plasmid p1896, which contains the promoter region of the leptin gene between -1951 and 1887 bp, and a potencial mutation site at the binding sites with Sp1.In the transsient transfections with this plasmid we observed that with exogenous Sp1 and HEGO, the leptin expression suffer a decrease, which could suggest the involvement of Sp1 in this expression.
