Ourlaboratorystudiestheregulationofpre-mRNAalternativesplicingbysignaltransductionpathways.WehavereportedthatAktactivationregulatestheactivityoftwosplicingfactorsoftheSRproteinfamily(SRSF1andSRSF7),alteringdifferentstepsalongmRNAmetabolism:alternativesplicingandtranslation.WealsorevealedAktasanSRproteinkinasecapableofphosphorylatingtheseSRproteins.WefoundthattheSRproteinSRSF1enhancesSUMOconjugationtoseveraltargetproteins,includingAkt.Here,thefurtherexploredthisunexpectedpost-translationalmodificationofAkt.BysitedirectedmutagenesiswemappedSUMOconjugationsiteswithinAkt.WefoundthatAktSUMOylationisrequiredforitsregulatoryroleonfibronectinandBcl-xaltrnativesplicing.Consistentwiththepro-survivalandpro-proliferativeroleofthiskinase,weshowedthatitsSUMOylationfavorstheproductionoffibronectinmRNAisoformscharacteristicofhighlyproliferativetissuesandtumors,aswellasofanti-apoptoticBcl-xmRNAisoforms.WealsofoundthatSUMOylationofAktcontrolsitsfunctionasaregulatorofcellcycleprogression.ThesefindingsrevealanovellevelofregulationforAktfunction,openingnewareasofexplorationrelatedtothemolecularmechanismsdeterminingthattheactivationofasinglekinasecanleadtoavastdiversityofcellularresponses.
