A natural antioxidant, resveratrol, prevents biochemical cardiovascular changes in fructose-fed rats.

Resumen:
This study describes the effect of chronic administration of Resveratrol (RV), an antioxidant comprised in the polyphenol fraction of red wine, on lipid peroxidation and cardiovascular nitric oxide síntesis in an experimental model syndrome “X” (FFR). Males Sprague-Dawley  rats were divided in 4 groups and treated during 45 d: Control (C n=6) gavage of vehicle (ethanol 10%); C+RV (n=6): gavage of R(10mg/Kg/día); FFRn (n=6): 10% fructose in drinking water + vehicle; and  FFR + RV (n=6). Changes in systolic blood pressure (Δ SBP mmHg) were recorded and oral glucosa toleraance test (OGTT mmol/L.120 min) were performed. Ca/calmodulin-dependent nitric oxide cintaza (eNOS) activity was determined in homogenates from mesenterio  resistance arteries (AeNOS) and cardiac tissue (CeNOS) by conversión of 3H-arginine and 3H-citrulline aand related to protein content (dpm/mgprot/min). Plasmatic thiobarbituric acid-reactive substances (TBARS µmol/L) were determined as index of lipid peroxidation. Tables shows the data (mean ± sem), processed by ANOVA and Bonferroni post-test ( *p<0.05 vs C; °p<0.01 vs FFR). The FFR model was able tod induce glucosa intolerante, increase in SBP and lipid peroxidation, and reduce cardiovascular eNos activity. RV was effective in the prevention of tensional rise, oxidative stress and eNOS recuperation. Data suggest that natural antioxidant consumption could be able to prevent functional and biochemical cardiovascular changes associated to this pathology.
