Zinc deficiency is a widespread problem. Zn essential roles in many aspects of the metabolism (Vallee and Falchuk, 1993), includes the activity of more than 300 enzymes, the structure of many proteins, and control of genetic expression. Zn chronic deficiency modified lipid parameters, especially phospholipids and cholesterol (Gómez et al., 2002). The present study was conducted to investigate the effects of Zn deficiency on the expression of lipidic enzymes such as ACC, LPL, HMG-CoA R, GPAT and SREBP2 in rat lung. Wistar male rats (200 ± 20 g) were fed with AIN-93 diet (zinc-deficient, ZD) or with 30 mg Zn/ kg (Control, Co) during 2 months. Total RNA was isolated from lung by using TRIzol. 1 mg of RNA was transcribed to cDNA at 42°C using random hexamers as primers and RT-MMLV (Moloney Murine Leukemia Virus). Aliquots of 2 mL of cDNA were used in the amplifications by PCR using specific primers. Beta actin was used as an internal control. We detected a significant increase in ACC (p < 0.01), LPL (p < 0.005) and SREBP2 (p < 0.01). HMG-CoA R (p < 0.005) decreased significantly, while GPAT did not change. From the results of this study, it can be concluded that Zn deficiency produces numerous alterations on lipids metabolism in lung.
