Resumen:
As2O3 induce apoptosis of leukemia cells but is also toxic and carcinogenic. This work proposal?s involves a lipid-protein system usage as carrier of arsenical compounds to reduce undesirable effects. Carrier lipids used were DPPC, SPC and DOTAP in concentration range of 1-5mM. The carrier protein was Trx (Thiorredoxin), chosen by its ability to interact with arsenic(III). Arsenical compound transported is AA(III)-FITC, obtained from in novo synthesis of arsalinic acid (AA) and fluorescein isothiocyanate (FITC). The complex system was obtained by incubating O.N. Trx and arsenic-fluorescent compound AA(III)-FITC, next step is association with liposomes. Association efficiency is higher for positive formulations. Positive charge liposomes also have the advantage of keeping size even 30 days after sample preparation. Trx-AA(III)-FITC has been characterized, there has not been seen a significant decrease in the native structure or loss of activity. Is was also analyzed in terms of UV-VIS absorbance, fluorescence and As amount and found a direct relationship between them. Finally, we have tested the effectiveness of the system in HL60 cell line, and it was possible to established a differential induction of apoptosis as compared to control systems.
