Our previous studies have shown increased MMPs and impaired PPAR signaling in diabetic embryopathy. PPARs signaling is activated by its coactivator PGC-1 and by its ligands oleic acid and certain eicosanoids derived from arachidonic acid through the activity of desaturases. Aims/Methods: To analyze the effect of a 6% olive oil-supplemented diet (OL) (enriched in oleic acid) to control and diabetic rats from days 1 to 10.5 of pregnancy on decidual PGC-1 and desaturase expression (PCR) and embryonic and decidual MMPs activity (zymography). We found reduced decidual PGC-1 and Ä5 and Ä6 desaturase expression in the diabetic group and that OL treatment increased PGC-1 expression. MMPs overactivity in embryos and decidua from diabetic rats was prevented by OL treatments. Conclusion: Maternal diabetes alters regulators of PPAR signaling in the decidua, and OL treatment improves both PPARs signaling and prevents MMPs overactivities in embryos and decidua from diabetic rats.
