Here we found a decrease onthe miR-203 expression which is coincident with an increase of its putativetargets, Phd12 and Snail2, during the initiation of neural crest cells (NCC) EMT.Bisulfite sequencing further reveals that this repression is epigeneticallymediated by DNA methylation on regulatory regions of miR-203 locus.Overexpression of miR-203 inhibits NCC migration, whereas functional inhibitionby using a ?sponge? vector promotes premature NC migration. Results from usingsensor vectors revealed that miR-203 can interacts with 3?UTRs for both PHD12and Snail2 genes. These finding demonstrated the in vivo existence offunctional epigenetic-microRNA regulatory network playing a significant role inthe development of NCC.
