Hairy root cultures are a useful system to study the contribution of roots as isolated organs in the response to different kinds of stress. We have use these cultures to analyze the effect of salt treatment on the tissue structure and to compare it with the response of normal roots. First we have analyzed the anatomy of both types of roots trough the observation under the microscope of slices from tissues included in paraffin. Tomato hairy roots showed the typical primary structure of roots from tomato plants, except for the presence of an exodermis, which was absent in roots of plants grown on vermiculite. When tomato hairy roots were grown on MS medium supplemented with NaCl (100 mM) or KCl (110 mM) and manitol (200 mM), tissues have shown better macroscopic aspect than the control cultures obtained in MS medium without supplements. We also observed less suberin deposition in cell walls of treated cultures. These cultures have shown secretion to the culture medium of a basic peroxidase isoenzyme, whose biochemical properties suggested that it is probably involved in the lignosuberization process.
