RAPID, NONGENOMIC ACTION OF PROGESTERONE ON CILLIAR MOVEMENT IN EPITHELIAL CELLS OF PIG OVIDUCT. Luisa Peralta, Mariela Roldán and Dora Miceli.  Dpto Biol. Argentina.   Our previous studies showed that progesterone (P) bind to specific sites on the membrane of pig oviductal cells. These binding sites presents the following kinetics parameters: Ka = 1.42 X 108 M-1, Kd = 7.05 x 10-9 M and Bmax = 3250 fmol/mg Prot. In this communication we studied the steroid effect on cilliar movement. We worked with ampulla epithelial cells from oviduct in estrogenic phase, obtained by colagenase digestion (1 mg/ml) in DMEM-Ham's F12 media. We followed the effect of P, mifepristone (M), corticosterone (C) and 17-Estradiol (E) during 5 min in an image analyzer (Macintosh, Image 1.45). The results show: 1) P (1 M) inhibited drastically the cilliar movement. After 30 sec we only observed a smooth movement. 2) M, C and E have not effect. 3) P + M (1 M) did not show effect either, indicating that M blocked the P effect. We confirm with competition assay that M displace P from the binding sites present in the plasmatic membrane IC50 = 1945 nM).
