A detailed biochemical characterization of the ooplasm wascarried out after elimination of the interference of theglycoproteins of the zona pellucida by both spectral subtractionand enzymatic degradation. The spectra thus obtainedallowed the identification of new spectral markers suitable todistinguish between immature and in vitro matured oocytes aswell as the observation of several weak bands correspondingto the less abundant components such as carbohydrates andpolynucleotides. This is the first study by Raman spectroscopyof bovine oocytes without the zona pellucida and constitutesthe basis for future assessments of structural/maturationaldeficiencies that could compromise the success of fertilization.
