Resumen:
The non-specific stimulation of the immune system is an interestingstrategy for the prophylaxis and early treatment of infections in circumstanceswhere there is a lack of specific treatments such as vaccinesavailable. This work aimed to evaluate different formulationsas non-specific immunostimulants in a murine model. The formulationswere based on an ODN-CpG combined with 4 gemini-type lipopeptides: AG2-C16, AG2-C18, AG10-C16 and AG10-C18. Theoptimal gemini:ODN-CpG ratio to achieve the ODN-CpG encapsulationwas determined using agarose electrophoresis. The immunostimulatorycapacity was studied in vitro and in vivo by qPCR.Total splenocytes collected from BALB/c mice, without prior treatment,were stimulated in vitro with the formulations or the individualcomponents. No stimulation was used as basal condition (control).Formulations based on AG2-C16 were evaluated in vivo using BALB/c mice immunized with 3 doses. Control group was immunizedwith buffer. Lymph nodes were harvested 72hs after last dose forRNA purification. In both studies, the expression levels of thesegenes were measured: IFN-γ, TNF-α, IL-6 and CD-80.
