Impaired microRNA 21 and prolactin receptor levels in the decidua of the prepuberal offspring of diabetic rats are prevented by maternal diets enriched in olive oil. OBJECTIVESDiabetes mellitus is a metabolic pathology that programs offspring´s diseases. We previously found reduced prolactin levels in the decidua of the offspring of diabetic rats, an alteration prevented by a maternal diet enriched in olive oil. MicroRNA 21 (miR-21) is a microRNA that regulates cell proliferation and decidualization and PPARdelta a PPAR isotype relevant in decidualization. Here, we aim to evaluate the levels of miR-21, PPARδ and the prolactin receptor PRLR in the decidualized uteri of prepuberal offspring of diabetic rats fed or not with an olive oil-enriched diet.METHODSA mild pregestational diabetic rat model was induced in F0 females by neonatal administration of streptozotocin (90 mg/kg sc). Control and diabetic females were mated with healthy males and received a 6% olive oil-enriched diet or a standard diet from day 1 of pregnancy until parturition. The offspring were fed a standard diet and the uteri of the female offspring (F1) were evaluated on postnatal day 30, after induction of decidualization with PMSG (50 UI) and hCG (50 UI). MiR-21, was evaluated by RT-qPCR and PPARδ and PRLR were evaluated by Western Blot.
