TPGS based 99mTC nano radiopharmaceuticals: potential application in breast cancer diagnosis.

Resumen:
There is only one useful SPECT radiopharmaceutical (99mTc-sestamibi)for breast cancer diagnosis and follow up and its utility carrieswith some limitations. Our aim was to evaluate the performance ofthree micellar 99mTc nano radiopharmaceuticals (i-TPGS; ii- TPGSand Soluplus®; iii- TPGS and Soluplus® with glucose) as radioisotopicimaging probes. Radiolabeling was performed by directmethod and a syngeneic orthotopic model of BALB/c mice injectedwith 4T1 cells, was used for scintigraphic diagnosis evaluationwith a gamma camera. The uptakes visualized in the images werevalidated by biological distribution measuring radioactivity accumulationin organs of interesr by Solid Scintillation Counter. Nano radiopharmaceuticalssizes were measured by Dynamic Light Scatteringat 25° and 37°C. TPGS and Soluplus® radiopharmaceutical wasthe only one that allowed scintigraphic visualization of the tumor.Nano radiopharmaceuticals sizes increased at 37°C for system iiand iii (more drastically for the latter). Biological distribution studiesshowed uptake values that backed up scintigraphic results for eachradiopharmaceutical assayed. In conclusion, mixed system of TPGSand Soluplus® stands as a potential candidate to be used in breast imaging diagnosis.
