In recentyears,speciallydesignedpatchescontainingbetaemittershavebeendevelopedforcontact brachytherapyofskinlesions.Theaimofthepresentworkwastoevaluatethebiologicaleffectsofthe 32P-patchontheskinofSencarmiceasaresultofabrachytherapytreatment.Forthispurpose,a 32P-patch waspreparedwithChromic 32P-phosphateandsiliconeandtheclassicalmodeloftwo-stageskin carcinogenesis wasreproducedinSencarmice.Animalsweredividedinsixgroups.Fourgroups receivedthecontactbrachytherapytreatmentsusingaschemeofasinglesessionof40and60Gy(SD40 and SD60)andaschemeoftwosessionsof40and60Gyeach(FD40andFD60).Theothertwogroups wereusedascontrolsofthesingle(CSD)andthefractionated(CFD)treatments.Radiationdoseswere estimated withequationsderivedfromtheMIRDDOSEscheme,andbiologicallyeffectivedoses(BED) werecalculatedaccordingtoequationsderivedfromthelinear-quadraticmodel.Theendpointto evaluatethetreatmentseffectswastumorsizeafterafollow-upperiodof44days.Finally,animalswere sacrificedinordertogetsamplesofalltumorsforhistologicalanalysisandPCNAstaining.Erythema, dermatitisandskinulcerationdevelopedinalmostalltreatedanimals,buttheygraduallyhealedwith regenerationoftissueduringthefollow-upperiod.RadiationeffectsontheskinofSD40,SD60,FD40 and FD60showedasignificantreductionofthetumorsizewithregardtocontrols,independentlyofthe scheme andtheradiationdoseconsidered.PCNAstainingscoresofcontrolgroupswerehigherthanfor treatedgroups,independentlyoftheschemeandtheradiationdoseconsidered.Thisradioactive 32P-silicone-patchwhichiseasytoprepareanduseinthetreatmentofskindiseases,seemspromisingasa radioactivedeviceforclinicaluse.
