Resumen:
The aim of this chapter was provide tools and protocols that should be followed in order to develop a successful research on soil bioremediation. We propose a step-by-step guidance towards the study of a contaminated site as a possible target for bioremediation. Some tips regarding the selection of the target site were provided. General protocols and international norms on sampling were taken into account, in order to maintain the purity of the samples and the correct management for their later treatment at the laboratory. Microbial isolation and their abilities for bioremediation vary according to the nature of the sample and as a consequence different methodologies were proposed. Assays developed on cultivable microorganisms to select those able to act on the contaminant of interest were also described. When the selection was finished, the analysis to establish which mechanisms are used to interact with the contaminant was described. Finally, we were focus on the use of molecular biology, such as proteomics and genetics to evaluate how microorganisms react towards the contaminant.This chapter includes specific highlights on the main issues to take into account when starting a research that aims towards soil bioremediation, so we expect it is useful for many researchers regarding this topic.
