Employing the model of carcinogen-induced tumors in SENCAR mice, we assessed the efficacy of ALA-PDT on papillomas, keratoacanthomas and in situ and invasive skin carcinomas. ALA was applied topically dissolved in different vehicles: saline lotion, ethanolic lotion, water in oil emulsion and oil in water emulsion. Lotions were the vehicles that induced higher porphyrin accumulation in papillomas. The invasive carcinomas responded better to topical ALA-PDT. The effect of PDT on papillomas was sligher, probably due to the lack of light penetration through the extense keratin layer. Vacuolization and cytoplasm condensation, nuclear pyknosis and nucleus disintegration were the most frequent cellular alterations. Vascular alterations such as congestion and hemorrhage were also seen in PDT-treated tumors. These findings may have great relevance in the treatment of skin cancers and probably for cutaneous T-cell lymphomas.
