Glycoproteomics emerge as the fusion field between proteomics and glycomics, pursuing the study of biological systems at a defined condition, handling the analysis of glycoproteins, including glycan structures, protein identity, glycosylation sites and level of occupancy. Our group has been focused on the structural study of glycoconjugates from unicellular to higher organism with the aim of revealing biological processes and functionality. The first steps on our research were supported with traditional chemical and enzymatic methodology and tools like HPAEC-PAD and MALDI-TOF MS. Lately, the acquisition of a nanoHPLC-ESI-Orbitrap, enabled us to speed up deeper glycoproteomics approaches from single purified proteins to samples that are more complex. I will show some of our recent studies on the structural determination of glycoproteins obtained from different strains of bacteria. This type of analysis allows us to find out in a purified protein both N- and O- glycosylation along with their respective glycan structures. I will focus in the workflow used to achieve our research for example the analysis in the positive and negative polarities to assure the assignments. I will also show that sugar components that have not been expected may be detected in this type of analysis and confirmed after nanoHPLC-ESI-Orbitrap analysis of the released oligosaccharide.
