De Robertis, Facultad de Medicina, Universidad de Buenos Aires firstname.lastname@example.org NMDA receptor (NMDAR) is involved in synaptic plasticity, learning, memory and neurological diseases like epilepsia and it is the major mediator of excitotoxicity. NMDARs are heteromeric complexes formed by the subunit NR1 (essential) and one or more NR2 subtype (AD). NR2A-containing NMDAR may be playing a crucial role in epileptic disorders. In this study we focused on NR2A subunit expression in hippocampus after repetitive seizures induced by 3-mercaptopropionic acid (MP) and the effect of adenosine analogue, cyclopentyladenosine alone (CPA) or previous to MP (CPA+MP). Lots of Wistar rats (200250 g) were administered during 4 days: a) MP 45 mg/kg; b) CPA 2 mg/kg; c) CPA+MP; d) Control (C). Hippocampus was homogenized at 5th day. Western blot assays was performed on the whole hippocampal tissue showing a significant increase in NR2A expression (17.5%, P<0.001) with respect to control. A tendency to reduce NR2A expression (7%) after CPA administration was observed. CPA previous MP (CPA+MP) showed a reduction of MP effect, recovering to normal values. NR2A immunoassays showed a staining of hippocampal pyramidal layer. This results suggest a citotoxic effect by MP-induced seizures and a protective effect by adenosine analogue.
