Resumen:
A review of pharmaceutical and biopharmaceutical properties of polyelectrolyte-drug solid products (PE-F) is presented. Such products are obtained by acid base reaction of acid- and basic-polyelctrolytes such as Carbomer (CB), Alginic acid (AA), Carboxymethylcellulose (CMC) and Eudragit E 100 (EU) respectively with ionizable F or opposite charge. PE-F complexes are stable amorphous solids that in presence of aqueous fluids quickly swell generating a hydrogel. By compaction of such materials swellable polyelctrolyte matrices (MHPF) are obtained. Kinetics and rate of F delivery from MHPF can be modulated by changes in their composition to satisfy the design of extended delivery systems. Among the factors that control de delivery of F, the rate of swelling, complex relaxation-dissociation of ionic pairs and further diffusion of F, as well as hydrogel layer erosion, are primarily involved. The prevalent mechanism of kinetic control depends of each particular system. PE-F Complexes under the form of matrices or multiparticulated systems may play an important role in the design of extended drug delivery systems.
