DESMOGLEIN-4 DEFICIENCY IMPAIRS HUMORAL IMMUNITY INDUCED BY OVA INTRADERMAL ADMINISTRATION

Desmogleins (Dsg) are transmembraneproteins located in desmosomes and play a critical role in the cell-cell adhesionof epithelial cells. Dsg-4 deficiency is associated with sharp hair loss inhumans, mice, and rats. Recently, we have reported a clear link between Dsg-4deficiency, exacerbated inflammation, and psoriasis-like lesions afterimiquimod (IMQ) administration. However, the role of Dsg-4 in humoral immunityhas not been addressed. Therefore, this work aimed to study the impact of Dsg-4 deficiencyin OVA-specific IgG induction. For this purpose, OFA hr/hrdeficient in Dsg-4, and wild-type Sprague-Dawley (SD) rats were inoculatedintradermally with PBS-OVA. IMQ, or Vaseline (Vas), was administered topicallyafter four hours from OVA injection and for the following three days. Two weeks later, serum samples were obtained to determined OVA-specific IgGlevels by ELISA. Surprisingly, OFA rats displayedlower OVA-specific IgG levels compared to SD group (OFA 0.118±0.09 vs D0.745±0.171 t test, p<0.0001). Although, IMQadministration induced a decreased OVA-specific IgG levels in both groups (SD Vas0.745±0.171 vs SDIMQ 0.446±0.236, t test, p<0.001; OFAVas 0.118±0.095 vsOFAIMQ 0.043±0.027; t test; p<0.01). These results suggest thatdesmoglein-4 promotes adaptive immunity in response to OVA challenge. Althoughthe underlying mechanism remains unknown, Dsg-4 deficiency may be impairing thefirst immune response events such as antigen draining to lymphatic nodes,antigen presentation, and germinal center reaction.
