TheglucosetransporterGLUT4isdynamicallysortedandretainedintracellularly andredistributestotheplasmamembrane(PM)byinsulin-regu-lated vesiculartraffic[1_TD$DIF], or ?GLUT4 translocation?. Hereweemphasizerecentfindings inGLUT4translocationresearch.Theapplicationoftotalinternalreflection fluorescence microscopy(TIRFM)hasincreasedourunderstandingof insulin-regulatedeventsbeneaththePM,suchasvesicletetheringandmembrane fusion.Wedescriberecent findings onAkt-targetedRabGTPase-activating proteins(GAPs)(TBC1D1,TBC1D4,TBC1D13)anddownstream RabGTPases(Rab8a,Rab10,Rab13,Rab14,andtheireffectors)along withtheinputofRac1andactin filaments, molecularmotors[myosinVa(MyoVa), myosin1c(Myo1c),myosinIIA(MyoIIA)],andmembranefusionregulators (syntaxin4,munc18c,Doc2b).Collectivelythese findings revealnovel eventsininsulin-regulatedGLUT4traffic
