Recent studies have investigated the composition andfunctional effects of extracellular vesicles secreted by variety of cell types.However, the mechanisms underlying the impact of these vesicles onneurotransmission remain unclear. Here, we isolated extracellular vesiclessecreted by hippocampal neurons and found that they contain synapticvesicle-associated proteins, in particular the vesicular SNARE (solubleNSF-attachment protein receptor) synaptobrevin (also called VAMP). Using acombination of electrophysiology and live-fluorescence imaging, we demonstratethat this extracellular pool of synaptobrevins can rapidly integrate into thesynaptic vesicle cycle of host neurons via a CD81- dependent process andselectively augment inhibitory neurotransmission as well as specifically rescueneurotransmission in synapses deficient in synaptobrevin. These findingsuncover a novel means of interneuronal communication and functional couplingvia exchange of vesicular SNAREs.
